Exfoliation by protease. Instead of loosening cell bonds with acid or scraping them off with grit, an enzyme peel uses a proteolytic enzyme to digest the keratin proteins holding dead cells together. It is the mildest of the three routes and the one least dependent on getting pH exactly right.
What it is
A protease — most often derived from fruit or from bacterial fermentation — in a powder, gel or cream base. The enzyme cleaves peptide bonds in the desmosomal proteins that anchor corneocytes to one another. Loosened, the dead cells rinse away.
Because enzymes act on protein rather than on the whole surface, the effect is self-limiting: once the accessible dead protein is digested, the reaction slows. That is the safety margin the category is sold on and it is broadly real.

Read the ingredient list before crediting the enzyme. A large share of products sold as enzyme peels also contain glycolic or lactic acid, and the acid is frequently the more active component.
Powder formats and the dust problem
Water-free powder formats exist because enzymes need water to work and are unstable in it — keeping them dry until the moment of use is the only way to guarantee activity. The trade-off is airborne dust. Papain, bromelain and subtilisin are all documented respiratory sensitisers; occupational asthma from proteases is well established in the detergent industry. Mix in the palm, over a sink, without inhaling, and this is a non-issue. Puffing powder around a bathroom repeatedly is not a good habit.
Who it suits
Reactive, rosacea-prone, thin or acid-intolerant skin, and anyone using a Retinoid Treatment who still wants a smoothing step. It is a weak choice for genuine congestion — salicylic acid reaches inside the pore and an enzyme does not — and for anyone chasing visible resurfacing.
Weekly is a reasonable ceiling. Leave on for the stated time and no longer; extra minutes buy irritation, not results.
Where it sits in the rules
Cosmetic only in the US — FDA primary category (14) — with exfoliating and smoothing claims. No monograph applies. Under MoCRA the responsible person must hold safety substantiation, and for a known sensitiser the sensible file includes justification of the enzyme concentration.
In the EU the product is notified through the CPNP and assessed in the product information file; the sensitisation potential of proteases is exactly the sort of thing the safety assessor is required to address. Neither regime sets a numeric limit on papain or bromelain in cosmetics.
Shelf life
Enzymes are the least stable actives in this catalogue. Heat, moisture and time all denature them, and a denatured protease is inert rather than dangerous — the product simply stops working, silently. Anhydrous powders hold activity longest. Twelve months after opening, kept dry and out of the sun, and treat any clumped powder as spent.
